centrifugationĀ  fromĀ  homogenatesĀ  ofĀ  C3H mouseĀ  liver.
OverĀ  80Ā  perĀ  centĀ  ofĀ  theĀ  totalĀ  isocitricĀ  dehydrogenaseĀ  activityĀ  ofĀ  the
homogenatesĀ  was recoveredĀ  inĀ  aĀ  fractionĀ  containingĀ  theĀ  solubleĀ  material
ofĀ  theĀ  cytoplasmĀ  ofĀ  theĀ  liverĀ  cell.Ā  OfĀ  theĀ  particulateĀ  fractions,Ā  nuclei andĀ  submicroscopicĀ  particlesĀ  showedĀ  veryĀ  littleĀ  activity,Ā  whereasĀ  mitochondriaĀ  containedĀ  approximatelyĀ  12Ā  perĀ  centĀ  ofĀ  theĀ  total.Ā  ResultsĀ  obtainedĀ  withĀ  hypertonicĀ  (0.88Ā  M)Ā  sucroseĀ  asĀ  theĀ  mediumĀ  wereĀ  essentiallyĀ  theĀ  same as thoseĀ  obtainedĀ  withĀ  isotonicĀ  (0.25Ā  M)Ā  sucrose.
OfĀ  theĀ  totalĀ  TPN-cytochromeĀ  c reductaseĀ  activityĀ  ofĀ  theĀ  homogenates,
overĀ  90Ā  perĀ  centĀ  wasĀ  recoveredĀ  inĀ  theĀ  particulateĀ  fractions,Ā  49Ā  perĀ  cent beingĀ  presentĀ  inĀ  mitochondriaĀ  and 36 perĀ  centĀ  inĀ  submicroscopicĀ  particles. UnderĀ  theĀ  conditionsĀ  ofĀ  theĀ  methodsĀ  ofĀ  assay,Ā  theĀ  specificĀ  TPN-cytochromeĀ  c reductaseĀ  activityĀ  ofĀ  mouseĀ  liverĀ  wasĀ  foundĀ  toĀ  beĀ  muchĀ  lowerĀ  thanĀ  valuesĀ  previouslyĀ  reportedĀ  forĀ  DPN-cytochromeĀ  c reductase.
SomeĀ  ofĀ  theĀ  difficultiesĀ  involvedĀ  inĀ  theĀ  interpretationĀ  ofĀ  dataĀ  obtainedĀ  withĀ  theĀ  cellĀ  fractionationĀ  techniqueĀ  areĀ  presented,Ā  andĀ  itĀ  isĀ  concludedĀ  thatĀ  severalĀ  definiteĀ  conditionsĀ  mustĀ  beĀ  fulfilledĀ  beforeĀ  aĀ  biochemicalĀ  propertyĀ  canĀ  beĀ  ascribedĀ  toĀ  aĀ  givenĀ  structuralĀ  componentĀ  ofĀ  theĀ  cell.
